SD Rat Aortic Smooth Muscle Cells
Introduction
Smooth muscle cells can be classified into vascular and visceral types based on their distribution and characteristics. Vascular smooth muscle cells (VSMCs), which reside in the tunica media of blood vessel walls, play a pivotal role in maintaining structural integrity and regulating vasomotor tone.
Vascular smooth muscle cells serve as important cellular models and tools for studying pulmonary heart disease, hypertension, cardiovascular and cerebrovascular atherosclerosis, diabetic vasculopathy, and vascular tissue engineering. Among various vascular sources, aortic smooth muscle cells are the most widely used due to their robust contractile phenotype.
OriCell™ SD Rat Aortic Smooth Muscle Cells are isolated from the aortic tissue of adult Sprague-Dawley rats via a combined trypsin-collagenase digestion method followed by differential adherence. The cells are subsequently purified and cultured in a specialized smooth muscle cell medium to ensure optimal growth and phenotype maintenance.

General Handing Principles
- Maintain strict aseptic technique. Ensure complete sterility throughout all procedures, particularly within the laminar flow hood and incubator.
- Follow standardized protocols. Adhere strictly to the product manual. Implement rigorous control over experimental variables and include appropriate parallel controls.
- Use high-quality consumables and reagents. This product requires culture vessels suitable for adherent cell growth, and the reuse of these vessels is not recommended. The reagents used must be validated for reliability, cell compatibility, and batch-to-batch consistency.
- Prioritize low-passage cells. Since OriCell™ SD Rat Aortic Smooth Muscle Cells have limited ability to proliferate in vitro (typically up to 2 passages), we recommend using low-passage cells for research applications.
- Optimize seeding density and subculture. The recommended seeding density for OriCell™ SD Rat Aortic Smooth Muscle Cells is (2.5–4) × 10⁴ viable cells/cm². Since cell growth is highly dependent on donor characteristics and culture conditions, we recommend adjusting the split ratios based on the actual performance of each specific lot and passage.
